ABSTRACT

OBJECTIVE To investigate the mechanism of Mayuan tongbian zhitong decoction on improving slow-transmission constipation(STC)in rats by regulating AMP activated protein kinase (AMPK)/endothelial nitric oxide synthase (eNOS)signaling pathway. METHODS The rats were randomly divided into blank group ,model group ,Mayuan tongbian zhitong decoction low-dose,medium-dose and high-dose groups (6,12,18 mg/kg),with 10 rats in each group. Except for blank group ,other groups were given Compound diphenoxylate suspension to induce STC model. After modeling ,blank group and model group were given normal saline intragastrically ,and Mayuan tongbian zhitong decoction groups were given relevant medicine intragastrically , once a day ,for consecutive 2 weeks. The number of feces and water content of feces in each group were observed before and after treatment;the carbon powder propulsion rate of rats in each group was calculated ;the pathological structure of colon in each group was observed ;the levels of nitric oxide (NO)and nitric oxide synthase (NOS)in colon tissues of rats in each group were detected;the expressions of AMPK ,eNOS,mammalian target of rapamycin (mTOR),tuberous sclerosis complex 1(Tsc-1), Tsc-2 and eukaryotic promoter 4E binding protein (4ebp)were also detected. The active ingredients of Cannabis sativa ,Citrus aurantium and Rehmannia glutinosa were screened from Mayuan tongbian zhitong decoction. The active ingredients with high Degree value were docked with AMPK and eNOS ,to verify the interaction. RESULTS Compared with before treatment ,the number and water content of feces were increased significantly in Mayuan tongbian zhitong decoction groups (P<0.05). Compared with blank group ,carbon powder propulsion rate of model group was decreased significantly (P<0.05); colonic structure was disordered ,and a large number of inflammatory cells were seen in submucosa ;the levels of NO and NOS in colon tissue as well as the protein expressions of AMPK ,eNOS,mTOR,Tsc-1,Tsc-2 and 4ebp were increased significantly (P<0.05). Compared with model group ,above indexes of Mayuan tongbian zhitong decoction groups (except for NOS in low-dose group )were reserved significantly (P<0.05). In the molecular docking experiment ,the active components with the highest Degree values in C. sativa ,C. aurantium and R. glutinosa were(Z)-3-(4-hydroxy-3-methoxy-phenyl)-N-[2-(4-hydroxyphenyl)ethyl] acrylamide,nobiletin and stigmasterol. The binding energies of AMPK with these three components were -5.15,-4.61 and -4.83 kJ/mol,the binding energies of eNOS with these three components were -6.11,-5.40 and -5.91 kJ/mol. The conformations of these three compounds with AMPK and eNOS were stable and their binding activities were high. CONCLUSIONS Mayuan tongbian zhitong decoction can improve the constipation symptoms and intestinal function in STC model rats ,and its specific mechanism may be related to the inhibition of AMPK/eNOS signaling pathway.

ABSTRACT

Objective:To examine the expression of transforming growth factor I(TGF-β1) and bone morphogenetic protein-9 (BMP-9) in human nonunion tissues,and to evaluate the clinical significance.Methods:The number of hypertrophic nonunion tissue samples and atrophic nonunion tissue samples were collected from Department of Orthopedics,the Second Xiangya Hospital of Central South University and Suzhou Kowloon Hospital Affiliated to School of Medicine of Shanghai Jiao Tong University between 2010 and 2014.Semi-quantification of SP immunohistochemical method and pathological image analysis software IPP6.0 were used to analyze the expression of TGF-β1 and BMP-9.Nonunion type,patients' age and nonunion time were statistical analyzed.Results:The absorbance values of TGF-β1 and BMP-9 in the hypertrophic nonunion tissues were 0.3236±0.0390 and 0.1337±0.0400,respectively;while the absorbance values of TGF-β1 and BMP-9 in the atrophic nonunion tissues were 0.3191±0.0369 and 0.1373±0.0423,respectively,with no significant difference between the two types of tissues (both P>0.05).There was also no significant difference in patients' age and bone nonunion time between them (all P>0.05).Conclusion:There is no significant difference in osteogenic potential between the hypertrophic nonunion tissues and the atrophic nonunion tissues.

ABSTRACT

To analyze and evaluate the clinical efficacy of the combination of locking plate and anti-osteoporosis drug for the patients with senior osteoporotic proximal humerus fractures. 120 patients with senior osteoporotic proximal humerus fractures were selected as research objects. According to the differences of treatment options, they were divided into two groups, i.e., 60 patients in observation group accepting the treatment of combination of locking plate and antiosteoporosis drug, the other 60 patients in the control group accepting simple locking plate treatment. The treatment effect and final result between the two groups were compared. The follow-up result showed that the average fracture healing time of observation group was [94.5 +/- 4.2] days, with excellent treatment rate reaching to 93.33% [56/60]; the average fracture healing time of control group was [116.5 +/- 3.8] days, with excellent treatment rate of 75% [45/60]. The intergroup different was of statistical significance, wherein p<0.05. The treatment efficacy of the combination of locking plate and anti-osteoporosis drug for senior osteoporotic proximal humeral fractures is faster and more precise, which is worth being applied in clinical practice

ABSTRACT

<p><b>OBJECTIVE</b>To explore the molecular basis for a CD36 deficiency individual and distribution of CD36 gene mutation in Guangxi population.</p><p><b>METHODS</b>A female individual was studied. CD36 phenotype was detected by monoclonal antibody immobilization of platelet antigens assay (MAIPA) and flow cytometry (FCM). The coding regions of the CD36 gene were sequenced. A DNA-based polymerase chain reaction-sequence specific primer (PCR-SSP) assay was used to verify the identified mutation. Cell lines expressing the mutant and wild-type CD36[CD36(MT) and CD36(WT)] were established, with the expression of CD36 determined by Western blotting. The distribution of CD36 gene mutation was investigated among 1010 unrelated individuals with the PCR-SSP assay.</p><p><b>RESULTS</b>Both MAIPA and FCM assays showed that the patient had type II CD36 deficiency. DNA sequencing showed that she has carried a heterozygous mutation T538C (Trp180Arg) in the exon 6 of CD36. Sequencing of cDNA clone confirmed that there was a nucleotide substitution at position 538 (538T>C). Western blotting also confirmed that the CD36 did not express on the CD36(MT) cell line that expressed the 538C mutant, but did express on the CD36(WT) cell line. The novel CD36 mutation T538C was further verified with 100% concordance of genotyping results by DNA-based PCR-SSP assay and 1010 unrelated individuals. No CD36 538C allele was detected among the 1010 individuals.</p><p><b>CONCLUSION</b>This study has identified a novel CD36 mutation T538C(Trp180Arg)(GenBank: HM217022.1), and established a genotyping method for the novel sequence-specific primer PCR. The novel mutation is rare in Guangxi and can cause type II CD36 deficiency.</p>

ABSTRACT

Objective To investigate gene polymorphism of Kell blood group in different Zhuang population from Guangxi region .Methods The genotypes of Kell blood group of 1025 non‐related individuals in different areas of Guangxi Zhuang popula‐tion were analyzed by PCR‐SSP .Results The Kell antigen in all individuals was homozygous ,the gene frequency of K and Jsa was 0 ,while that of k and Jsb was 1 .000 .Conclusion The distribution characteristic of Kell blood group in Guangxi Zhuang population was monomorphism ,which was similar to other Chinese population reported by literatures .

ABSTRACT

Objective Study on the distribution of auto‐antibodies against platelet in adults and children with primary immune thrombocytopenia(ITP) .Methods Platelet auto‐antibodies were detected in 83 ITP patients and 58 non‐ITP patients by enzyme linked immunosorbent assay (PAKAUTO kit) .Anti‐GP Ⅱ b/Ⅲ a ,GP Ⅰ b/Ⅸ and GP Ⅰ a/Ⅱ a auto‐antibodies were analyzed for 46 a‐dults and 37 children with ITP .Results The positive rate of autoantibody was 66 .27% in ITP patients ,which was much higher than that of non‐ITP patients ,the difference was significant(P 0 .05) ,but the morbidity of ITP in females was higher than that of males in both of the two groups ,the difference was significant(P 0 .05) ,the main of them was resistance against GP Ⅱ b/Ⅲ a and GP Ⅰ b/Ⅸ antibodies .Conclusion The detection of platelet auto‐antibodies could distinguish ITP and non‐ITP patients ,and GP Ⅰ a/Ⅱ a has an important reference value about therapy .

ABSTRACT

Objective To investigate the distribution situation of ABO and Rh blood groups among Zhuang population in Nanning area to guide the scientific voluntary blood donor recruitment and rational storage of blood in this area.Methods 2 052 blood samples from Zhuang population in Nanning area were performed the identification of ABO and Rh blood groups.Whether the gene distributions conforming to the Hardy-Weinberg equilibrium principle was assessed by using the Chi-square test.The data was compared with the distribution characteristics in other areas or other minorities.Results The distribution characteristics of ABO blood groups in Nanning area were O > B> A > AB.Blood group O was highest (46.78%)and blood group AB was lowest (4.34%).The gene frequencies of A,B and O were 0.131 9,0.181 5 and 0.686 6,respectively.The blood groups distribution con-formed to the Hardy-Weinberg equilibrium principle;the RhD positive proportion(RhD+ )in Zhuang population was 99.90%,while the RhD negative (RhD- )proportion was 0.10%,which was lower than that in Han population.No phenotypes of ccdE,CcdE, CCdee,CCdE,CCDEE and ccDee were observed in this study.Conclusion The distribution of the ABO and Rh blood groups among Zhuang population in Nanning still maintains their own exclusive characteristics.The extremely low proportion of RhD- will bring about serious difficulties to the patients with long term blood transfusion.

ABSTRACT

BACKGROUND:Peri-fracture nerve injury can inhibit osteoclast activity and promote early fracture healing. OBJECTIVE:To investigate dynamical y the effects of traumatic brain injury on the bone mineral density, microstructure, biomechanics property and bone metabolism in rat models of fractures. METHODS:Sixty-three male rats were randomly divided into three groups:sham group, simple fracture group and fracture combined with brain injury group. After 3, 6, and 3 months, the animals were sacrificed in batches under anesthesia, and then, the bones and serum specimens were used to detect the bone mineral density, microstructure, biomechanics property, serum cross-linked N-telopeptide of col agen type I and osteocalcin levels. RESULTS AND CONCLUSION:Compared with the simple fracture group, the fracture combined brain injury group had significantly increased bone mineral density of the proximal tibia, bone volume fraction of the cancel ous bone, trabecular thickness, cross-sectional area of tibial cortical bone and total area of the bone marrow, ultimate load and stress of the tibia, serum cross-linked N-telopeptide of col agen type I and osteocalcin levels at 3 and 6 weeks after modeling (P<0.05), but no differences in the above-mentioned indexes were found among the three groups at 3 months after modeling. These findings indicate that traumatic brain injury can increase the bone mineral density at the fracture site, improve bone microstructure and enhance biomechanical properties, thereby promoting bone healing and bone metabolism at the fracture site.

ABSTRACT

Objective To establish the platelet antigen panel cells and to apply them in the detection and identification of platelet alloantibody.Methods Human platelet antigen(HPA)1-16 genotyping from 1 500 un-related blood donors in Nanning area were performed by the polymerase chain reaction-sequence specific primers(PCR-SSP)technique,platelet antigen cells with O blood type were chosen to establish the panel cells of platelet antigen.The phenotype of the panel cells were verified by the reference sera from the 14th platelet immunology workshop of the International Society of Blood Transfusion(ISBT).And then these panel cells were used in clinic to detect the platelet alloantibody and the samples from the 14th and 15th platelet immunology workshop of ISBT.Re-sults Six platelet cells with consistent phenotype and genotypes and covering the HPA 1-5 and 1 5 systems were selected to estab-lish the platelet panel cells and successfully applied them in the clinical and scientific sample detection and identification.Conclusion Platelet antigen panel cells are established successfully,which provides the experimental basis for the diagnosis and research of platelet allogenic abnormal immunity diseases.

ABSTRACT

Objective:To discuss the application of multiple seasonal ARlMA model to data in time series and fit predictive model on workload of inpatient department.Methods:Parameter of model is estimated using conditional least squares method according to the data of workload of inpatient department in a hospital in Chongqing from 2002 to 2005.The structure is determined according to criteria of residual un-correlation and concision.ARIMA predictive model was fitted and the order of model was confirmed through Akaike's Information Criterion and Schwarz Bayesian Criterion.Results:Statistics assisted estimation of the model.The estimation of variance is 0.003 175,AIC=-94.245 1,SBC=-91.252 1.Using analysis of white-noise residual of mode1,fit of rich table show that the best estimated ARIMA model is(1-B)(1-B12)Zt=(1-0.543 02B)(1-B12)at.Conclusion:The model of ARIMA can be used to forecast for workload of inpatient department with high prediction precision of short-term time series.
